Among the antimicribial agents, the
4-quinolone class generates a considerable
interest since its discovery
in 1962 with nalidixic acid, and its
introduction for clinical use in 1963,
so marking the beginning of five
decades of quinolone development
and use. Ever since, significant progress
has been made in our understanding
of the molecular mechanisms
of action of quinolones against
pathogenic bacteria, the selection of
resistance among these microorganisms,
and the potency of each of
these compounds to induce toxic
and side effects in different treated
patients. This literature review, divided
into two articles, previously presented
the viewpoint of the bacteriologist
on this antibiotic family. This
second part, within the continuity of
the previous, will pay attention to the
pharmacodynamic and pharmacokinetic
properties of 4-quinolones,
the toxicity and the main side effects
reported and the main indications in
