Among the antimicribial agents,
the 4-quinolone class generates a
considerable interest since its discovery
in 1962 with nalidixic acid,
and its introduction for clinical use
in 1963, so marking the beginning
of five decades of quinolone development
and use. Ever since, significant
progress has been made in
our understanding of the molecular
mechanisms of action of quinolones
against pathogenic bacteria,
the selection of resistance among
these microorganisms, and the
potency of each of these compounds
to induce toxic and side
effects in different treated patients.
This literature review will be divided
into two articles that will present
the current state of pharmacological
and antimicrobial activity on
this antibiotic family. The first part
of this review will detail the relation
between the structure and the activity
of these drugs, the classification,
the activity spectrum and the
